Differential Metabolites in Chinese Autistic Children: A Multi-Center Study Based on Urinary 1H-NMR Metabolomics Analysis
نویسندگان
چکیده
Background: Autism spectrum disorder (ASD) is a group of early-onset neurodevelopmental disorders. However, there no valuable biomarker for the early diagnosis ASD. Our large-scale and multi-center study aims to identify metabolic variations between ASD healthy children investigate differential metabolites associated pathogenic mechanisms. Methods: One hundred seventeen autistic 119 were recruited from research centers 7 cities. Urine samples assayed by 1 H-NMR metabolomics analysis detect variations. Multivariate statistical analysis, including principal component (PCA), orthogonal projection latent structure discriminant (OPLS-DA), as well univariate used assess control groups. The further analyzed receiver operating characteristics (ROC) curve pathways analysis. Results: Compared with group, showed higher levels glycine, guanidinoacetic acid, creatine, hydroxyphenylacetylglycine, phenylacetylglycine, formate lower 3-aminoisobutanoic alanine, taurine, creatinine, hypoxanthine, N-methylnicotinamide. ROC relatively significant diagnostic values hypoxanthine [area under (AUC) = 0.657, 95% CI 0.588 0.726], creatinine (AUC 0.639, 0.569 0.709), creatine 0.623, 0.552 0.694), N-methylnicotinamide 0.595, 0.523 0.668), acid 0.574, 0.501 0.647) in group. Combining AUC reached 0.720 (95% 0.659 0.777). Significantly altered metabolite serine threonine metabolism, arginine proline taurine hypotaurine metabolism. Conclusions: Urinary amino significantly Amino might play important roles mechanisms

Objective: This research revealed the biochemical outcomes of metabolic dysregulation in serum associated with physiological sickness behavior following lipopolysaccharide (LPS)-induced neuroinflammation in rats, and treatment with Clinacanthus nutans (CN). Verification of 1H NMR analysis of the CN aqueous extract proved the existence of bioactive phytochemical constituents’ in extract. Materia...
متن کامل1H NMR metabolomics study of age profiling in children.
Metabolic profiling of urine provides a fingerprint of personalized endogenous metabolite markers that correlate to a number of factors such as gender, disease, diet, toxicity, medication, and age. It is important to study these factors individually, if possible to unravel their unique contributions. In this study, age-related metabolic changes in children of age 12 years and below were analyze...
متن کاملAssessment of 1H NMR-based metabolomics analysis for normalization of urinary metals against creatinine.
BACKGROUND Proton nuclear magnetic resonance (1H NMR, or NMR) spectroscopy and inductively coupled plasma-mass spectrometry (ICP-MS) are commonly used for metabolomics and metal analysis in urine samples. However, creatinine quantification by NMR for the purpose of normalization of urinary metals has not been validated. We assessed the validity of using NMR analysis for creatinine quantificatio...
متن کامل1H-NMR-based metabolomics study of cerebral infarction.
BACKGROUND AND PURPOSE Stroke is one of the leading causes of adult disability and death in developing countries. However, early diagnosis is difficult and no reliable biomarker is currently available. Thus, we applied a 1H-NMR metabolomics approach to investigate the altered metabolic pattern in plasma and urine from patients with cerebral infarctions and sought to identify metabolic biomarker...
